黄瓜再生体系和遗传转化的建立

摘    要:以黄瓜(Cucumis sativus L.)哈研3号为试验材料,采用组织培养技术系统探讨以子叶或子叶节为外植体时不定芽的诱导效率,最终确定以子叶节作为再生体系及遗传转化体系的试验材料;并系统探讨了不同PGRs浓度对不定芽诱导、不定芽伸长及生根阶段的影响;建立了哈研3号黄瓜的高频再生体系;并在子叶节根端膨大阶段进行了抗生素(Km)浓度的摸索,建立了较为适宜的遗传转化体系,为转基因工作打下坚实的基础。
