小鹅瘟病毒GD-06株的分离鉴定

摘    要:从广东某地死亡的、具有小鹅瘟典型病变的雏鹅肝、脾中分离到一株病毒。采用鹅胚增殖病毒,通过电镜观察到细小病毒样粒子;人工感染7日龄健康易感雏鹅,引起100%的发病和死亡,并具有典型小鹅瘟的临床症状和剖检特征。经琼脂扩散试验,用此株病毒感染的鹅胚液制备的小鹅瘟沉淀抗原能与小鹅瘟病毒阳性血清发生特异性反应。针对编码主要结构蛋白VP3的基因设计合成一对引物,扩增出与预期长度相符的特异性DNA片段。

Abstract:One wild strain of gosling plague virus(GPV) was isolated from the liver and spleen of affected young goose in GuangDong province.The virus were propagated in goose embryos,the typical gosling plague virus was observed by electronic microscope..At the age of 7 days ,the goose were artificially inoculated with virus sample and the pathological lesion were similar to the GPV natural infected cases.The GPV antigen from allantoic fluid of infected embryos could react with positive serum of gosling plague by agar gel precipitation.A pair of primers were designed and synthesized,which were directed to the gene code for VP3.The amplified DNA fragment was in accordance with the expected length.
