文冠果雄性可育性相关cDNA片段的克隆与序列分析

摘    要:针对文冠果单性雄花与两性花中存在的可育与败育两种不同类型雄蕊 ,为在分子水平上探讨雄性不育的机理 ,作者采用mRNA差别显示技术 ,分析始花期两种花药中基因的表达情况 .从雄花花药中初步筛选到N15和N2 42条与雄性可育性相关的cDNA片段 .同源性比较发现 ,N15片段与大豆质体rps12、rps7、16SrRNA、NADH脱氢酶基因共转录序列的同源性达 95 %,并与矮牵牛雄性可育系线粒体中的可育性相关基因结构吻合 ;N2 4片段的部分序列同拟南芥染色体 3中CHR3v0 7142 0 0 2基因序列的同源性为 80 %.该研究为进一步探讨育性相关基因调控文冠果育性的分子机理奠定了基础 .

Abstract:Aiming at the subject that two kinds of stamens, fertile and sterile, exist respectively in staminate and hermaphrodite flowers of Xanthoceras sorbifolia Bunge, the authors analyze the pattern of gene differential expression in the two kinds of early flowering anthers by means of DDRT--PCR so as to probe into the molecular mechanism of male sterility. Two fragments of cDNA, N15 and N24, correlated with male fertility were selected preliminarily from the specific fragments obtained in male flower anthers. Fragment N15, 542 bp in length, had 95% homology with soybean plastid DNA for rps12,rps7,16S rRNA,NADH dehydrogenase and ORF143; and was identical with the mitochondrial gene organization related to pollen fertility in petunia. Fragment N24 was 600 bp in length, its partial sequence showed 80% identity with the corresponding sequence of gene CHR3v07142002 locating in chromosome3 of Arabidopsis thaliana. This research lays a foundation for further study on the molecular mechanism of gene regulating male fertility or sterility in Xanthoceras sorbifolia Bunge.
