大麻种子萌发时总糖和还原糖含量的动态变化及其检测方法优化

摘    要:以工业大麻品种云麻1号和M641萌发过程中的种子为实验材料,对DNS法(3,5-二硝基水杨酸法)测定还原糖的条件进行了研究,并采用蒽酮法测定总糖含量,分析了大麻种子在萌发过程中总糖和还原糖的动态变化。结果显示,DNS法测定还原糖的最佳检测条件为:检测波长530 nm,DNS用量2 mL,显色时间7 min。在种子萌发过程中还原糖和总糖的含量表现出先下降后上升,但整体呈上升趋势,其中,云麻1号和M641的总糖含量总体升幅分别为158%和78%,还原糖含量总体升幅分别为671%和703%。本研究探明了大麻种子萌发过程中总糖和还原糖含量的变化,可为大麻种子萌发机理研究提供参考,同时也可为今后大麻植物及种子中的还原糖和总糖的测定提供借鉴。

Abstract:Dynamic changes of total sugar (by Anthrone method) and reducing sugar (by DNS method) in the process of hemp seed germination were investigated using seeds from two hemp (Cannabis sativa L.) varieties YunMa 1 and M641.The results showed that the optimum determination wavelength of DNS method was 530 nm,the proper volume of DNS reagent was 2.0 mL,and the optimal time for colour reaction was 7 min.The contents of total and reducing sugar increased distinctly after a slow declining in early stages of the germination process.The contents of total sugar of YunMa 1 and M641 increased by 158% and 78% respectively,and the reducing sugar increased by 671% and 703% respectively.The dynamic changes of total and reducing sugar in the germination process of hemp seeds revealed in this paper could provide reference for the research of germination mechanism and also for the determination of carbonhydrate of hemp seeds.
