野生甘蒙锦鸡儿的组织培养研究

摘    要:本试验采用甘蒙锦鸡儿(Caragana opulens Kom.)成熟种子为材料,利用组织培养的方法进行成熟胚的诱导,当胚成苗后进行增殖和生根培养,建立了一套野生甘蒙锦鸡儿的组培体系,为保护甘蒙锦鸡儿野生资源及其引种到城市园林建设中提供一定的借鉴作用。结果表明:试验前将种子用蒸馏水浸泡一晚效果较好;当灭菌时间为15 min时污染率最小,为37.61%;MS+6-BA 0.5 mg·L-1+NAA 0.05 mg·L-1培养基为最适宜成熟胚离体诱导的培养基;MS+6-BA 0.5mg·L-1+NAA 0.05 mg·L-1培养基为最适宜增殖的培养基;再生苗移植到1/2 MS+IAA 0.1 mg·L-1培养基上,植株健壮,生根率为63.16%;将生根培养的甘蒙锦鸡儿幼苗移栽到草炭土:珍珠岩:蛭石=6:3:1的混合基质中,成活率为90%。

Abstract:In this experiment,mature seeds of Caragana opulens Kom. were used as materials to induce mature embryos by tissue culture. After emergence of seedlings,shoot multiplication culture and rooting culture were made. In this test,a set of tissue culture system of wild C. opulens. was established to provide some reference for protecting wild resources of C. opulens. and its introduction to urban garden construction. The test results showed that it was better to soak the seeds in distilled water for one night before the experiment. The minimum contamination rate was 37.61% when the sterilization time was 15 minutes. MS + 6-BA 0.5 mg·L-1 + NAA 0.05 mg·L-1 medium is the most suitable medium for in vitro induction of mature embryos. MS+6-BA 0.5 mg·L-1+NAA 0.05 mg·L-1 medium was the most suitable medium for proliferation. When the regenerated seedlings were transplanted on 1/2 MS + IAA 0.1 mg·L-1 medium,the plant was robust,and the rooting rate was 63.16%. When the rooting seedlings of C. opulens. were transplanted into a mixed growth medium,in which the ratio of peat:perlite:vermiculite is 6:3:1,the survival rate was 90%.
