刺五加多糖提取纯化工艺的优化

摘    要:[目的]优化刺五加多糖的提取纯化工艺。[方法]以药用植物刺五加为材料,采用单因素试验和正交试验法对多糖提取、纯化的工艺进行优化。[结果]结果表明,热水浸提的最佳温度为90℃,浸提时间为150min,水料比为18:1,提取次数2次;醇沉温度为-20℃,时间36h,乙醇与浸提液比例为4:1,pH值为7时多糖产率最高。采用树脂动态吸附法筛选不同树脂去除多糖色素和蛋白的效果,结果显示,D941树脂最好。经树脂处理的多糖采用DEAE-Sepharose阴离子交换柱和Sepharose-G200层析柱进一步纯化,得到了纯度较高的As-1和As-2 2个多糖组分。[结论]该研究结果为进一步确定刺五加多糖的生化特性、生理功能奠定基础。

Studies on the Extraction and Purification Techniques of Polysaccharides from Acathopanax senticosus

Abstract:[Objective]The purpose of this research was to optimize extraction and purification techniques of Polysaccharides from Acathopanax senticosus.[Method] The extraction and purification conditions of polysaccharides from Acathopanax senticosus were studied by single factor tests and orthogonal design tests.[Result]The results indicated that the conditions of extraction and purification techniques of Polysaccharides from Acathopanax senticosus were as follows: the optimal water extraction conditions of polysaccharides were confirmed that the water temperature for extraction was 90 ℃ for 150 min,2 times extraction,and the ratio of material(g) to water(ml) was 1∶18.The optimal alcohol sedimentation conditions of polysaccharides were ascertained that the alcohol temperature was -20 ℃ for 36 h,the ratio of alcohol(ml) to water extraction portion(ml) was 4∶1,and the suitable pH value was 7.The effect compares of decolorization and deprotein from the crude polysaccharides among different resins suggested that the D941 resin was of the best effect on decolorization and deprotein.The polysaccharides were further purified by DEAE-Sepharose and Sephadex G-200 chromatogram column,then As-1 and As-2 of polysaccharides portion were obtained.[Conclusion]This study will help research the biochemical characteristic and physiological function of Acathopanax senticosus polysaccharide.
