山葡萄基因组DNA提取及RAPD鉴定

摘    要:以雌花山葡萄(73064)冷藏幼叶、香妃葡萄新鲜幼叶为试材,分别采用修改的CTAB法、高盐法、SDS法提取基因组DNA。结果表明,三种方法所提的DNA纯度及产率有差别,从综合结果看,以CTAB法为好,所得DNA不需经氯化艳梯度离心或柱层析,可直接用于RAPD分析。

Abstract:The extracts of high quality genomic DNA was extracted from the frozen leaves of V. amurensis Rupr. (■,73064) and the fresh leaves of Xiangfei(V. vinifera L. )by modified CTAB, medium with high salts and SDS method respectively. The purity and production were different among those 3 methods. According to the comprehensive results, the method of GB was the best (A260/A280 = 1. 7, production 57 ng' mg~(-1) fresh leaves). ms method didn' t need the liquid N and phenol extraction, and the DNA length was similar to λ DNA (48 kb) .TIs isolated DNA could be used directly for RAPD analysis without sedimentation in cesium chloride gradient or column chromatography. It's really suitable for great amount samples for RAPD analysis.
