柿离体繁殖研究

摘    要:以成年柿树休眠芽为外植体,研究了影响柿树离体繁殖的有关因素。结果表明:附加ZT0.5~2mg·1-1和IAA0.05~0.1mg·1-1或仅加ZT2mg·1-1的改良MS培养基对柿芽的增殖和生长最为适宜。在继代培养中,适当缩短每代培养时间可加快新梢的增殖速度。IAA诱导生根的效果优于IBA,用生长素溶液浸新梢基部后转入1/2MS中对生根更有利。在较低温度下,以腐殖土作基质,试管苗的移栽成活率可达88.6%。

Abstract:Dormant buds of persimmon trees were taken as explants for in vitro culture,some factors affecting in vitro propagation were studied. The results showed that modified MS medium supplemented with ZT 0.5 ~ 2mg. 1-1 and IAA0. 05 ~ 0. 10mg. 1-1, or ZT2mg.1-1 only were most suitable for the proliferation and growth of persimmon buds. In the proceeding of subculture,shortening times of subculture could accelerate the proliferation rate of shoot. The inducing rooting activity of IAA was better than that of IBA. Dipping the basis of shoots in the auxin solution before cultured it into 1/2 MS medium was better than adding auxin into the medium directly for rooting. Under lower temperature,using humus soil as the transplant medium,the survival rate of plantlet could be reached to 88.6%.
