妊娠失败牛外周血中干扰素刺激基因和细胞因子的表达分析

摘    要:为了探究牛人工授精后妊娠失败的原因,本研究分析了妊娠失败牛外周血干扰素刺激基因和细胞因子等的表达变化。试验选取11头健康状况良好、体况相近的未孕奶牛,采集人工授精后0d、18d和28d的血液样本,结合28d血清PAG检测和42d直肠触诊,对确定为妊娠失败牛的血液样本进行分析。结果表明,与0d相比,人工授精后18d血液RSAD2、MX1和RTP4 mRNA的表达均呈上调趋势,但无显著差异;18d时IL-2 mRNA的表达显著高于0d,IL-10和CCL8 mRNA的表达显著低于0d,IFN-γ、IL-4和CXCL10 mRNA的表达均呈上调趋势,但无显著差异。说明妊娠失败与胚胎发育早期母体免疫失调及妊娠识别能力不足有关。

Abstract:In order to investigate the reasons of pregnancy failure in cows after artificial insemination,the expression of interferon stimulated(ISGs)and cytokine in peripheral blood of non-pregnant cows were analyzed,so as to provide theoretical basis for the formulation of embryo death protection measures for cows after hybridization.11 unpregnant cows with good health and similar body condition were selected,blood samples were collected on 0d,18d and 28d after artificial insemination,and the blood samples of cows identified as pregnancy failure were analyzed by combining serum PAG detection on 28d and rectal palpation on 42d.The results showed that:compared with 0d,the mRNA expressions of RSAD2,MX1 and RTP4 in blood were up-regulated on 18d after artificial insemination,but without significant differences;the mRNA expression of IL-2 on 18d was significantly higher than 0d,however,mRNA expression of IL-10 and CCL8 were significantly lower than 0d;the mRNA expressions of IFN-γ,IL-4 and CXCL10 on 18d were up-regulated,while no significant differences were observed.It suggested that maternal immune disorders during early embryonic development and the weaker ability of pregnancy recognition may be related to pregnancy failure.
