BCO2基因多态性与麻黄鸡皮肤黄度的相关性研究

摘    要:试验旨在研究麻黄鸡皮肤黄度的遗传特性,以及β-胡萝卜素加氧酶2(BCO2)基因多态性与麻黄鸡皮肤黄度的关联性。选取310只80日龄商品代麻黄鸡,利用肤色仪对屠宰后鸡的胫部、腹脂以及5个不同躯干部位的颜色进行黄度值测定,并分析不同部位黄度值之间的相关性。同时筛查BCO2基因SNPs位点,鉴定个体基因型,分析基因多态性与麻黄鸡皮肤黄度的关联性。结果显示:(1)胫色与躯体皮肤黄度无相关,与腹脂黄度也无相关(P>0.05);腹脂黄度与躯体整体的皮肤黄度极显著相关,躯体5个不同部位的皮肤黄度相互之间具有极显著相关性(P<0.01);(2)BCO2的3个经典SNP(chr24:6264085、chr24:6273428、chr24:6287900)与试验群体的皮肤黄度无相关(P>0.05);(3)单倍型GGTTAA个体的前胸黄度b*值显著高于单倍型GACTGG(P<0.05),同时高于单倍型AATTGG和单倍型GACTGA个体。结果揭示了麻黄鸡胫色、腹脂颜色和表皮颜色相互之间的关系,发现BCO2基因上的一个单倍型与麻黄鸡皮肤黄度性状相关,为探讨其作为有效遗传标记的可行性提供了参考,为麻黄鸡的分子育种提供了依据。

Abstract:This experiment was to study the genetic characteristics of skin color in Partridge chicken, and analyzed the correlation between the polymorphism of BCO2 gene and skin yellowness traits in Partridge chicken. Three hundred and ten of 80-day-age commercial Partridge chicken were used as experiment materials. The yellowness of shank, abdominal fat and five different trunk parts of carcass were collected by using colorimeter, and correlation between the yellowness values of different skin parts was analyzed. The polymorphisms of BCO2 gene were screened to identify the individual genotype,and the correlation between gene polymorphism and skin yellowness value of Partridge chicken was also analyzed.The results showed that there was no significant correlation between shank skin yellowness and trunk skin yellowness,and the yellowness of abdominal fat was also not related to the shank skin yellowness(P>0.05).The abdominal fat yellowness was significantly related to the mean skin yellowness, and the shank skin yellowness from five different parts of the trunk was significantly related to each other(P<0.01). Three classical SNPs of BCO2, including chr24: 6264085, chr24: 6273428,chr24: 6287900, were not related to the skin yellowness in this experimental population. The yellowness value of precordial of haplotype GGTTAA was higher than that of haplotype GACTGG(P<0.05), and it was also higher than that of haplotype AATTGG and GACTGA. The results demonstrated that the relationship among shank skin color, abdominal fat color,and trunk skin color, and found a haplotype of BCO2 was correlated with the skin yellowness in Partridge chicken.These results not only advance our understanding of the genetic characteristics of chicken skin color, but also provided a reference for using a haplotype of BCO2 gene as a genetic marker for skin yellowness selection in Partridge chicken.
