一株高抗镉细菌KCd1的分离鉴定及其抗性基因初步研究

摘    要:从北京凉水河床底泥中分离得到一株能在含450 mg/L CdCl2的LB平板正常生长的高抗镉细菌,编号为KCd1。16S rDNA序列分析结果显示,该菌株与Bacillus cereusATCC 14579T的序列同源性达99%;结合脂肪酸测定与生理生化测定结果,确定该菌株在分类上属于Bacillus cereus。根据GenBank中已发表的抗镉细菌cadA序列设计了巢式引物,以菌株KCd1基因组DNA为模板,扩增得到约500 bp的PCR产物,将该PCR产物克隆于pGEM T-Easy载体,测序结果显示其大小为468 bp,与已报道的Staphylococcus haemolyticus的cadA基因对应序列相似性达到99%,由此可知本实验所获得到468 bp基因片段为cadA基因的一段保守序列。该菌株cadA全长基因的获取工作还有待于进一步展开。
