摘    要:[目的]为食品添加剂胭脂红的合理使用提供依据。[方法]蚕豆种子经浸种、催芽后,用不同浓度的食品添加剂胭脂红和分析纯胭脂红处理24h,以蒸馏水处理为对照,样品固定染色压片后镜检,计算根尖细胞的微核千分率和染色体畸变百分率。[结果]当胭脂红浓度为0.02和0.04g/L时,蚕豆初生根色泽鲜亮,与对照处理无差异。随着胭脂红处理浓度的增大,蚕豆根的颜色呈黄-褐-黑的变化趋势。2种胭脂红均可诱导蚕豆根尖细胞产生微核和染色体畸变,且微核率和染色体畸变率均随处理浓度的升高而增大,分析纯胭脂红诱导的微核率和染色体畸变率多低于食品添加剂胭脂红。[结论]过量使用胭脂红会对生物体产生毒害。

Influence of Coccinellin Concentration on Caryomitotic in Root Tip Cells of Broad Bean

Abstract:[Objective]The purpose was to supply basis for reasonable application of food additive coccinellin.[Method]After seed soaking and accelerating germination,the broad bean seeds were treated with food additive coccinellin at different concn.(concentration) and analytical pure coccinellin for 24 h,with distilled water treatment as CK,the samples were fixed,dyed and squashed for microscopic examination.The micronucleus permillage and chromosomal aberration percentage in root tip cells were calculated.[Result]When coccinellin concen.were 0.02 and 0.04 g/L resp.,the color of newly grown root of broad bean was bright and had no difference from that of CK.As the coccinellin concen.was increased,the color of broad bean root showed variation trend of yellow-brown-black.Both the 2 kinds of coccinellin could induce the root tip cells to generate micronucleus and chromosomal aberration and their micronucleus permillage and chromosomal aberration percentage were increased along with the ascending of coccinellin concen.Most of the micronucleus permillage and chromosomal aberration percentage in cells induced by analytical pure coccinellin were lower than that induced by food additive coccinellin.[Conclusion]The application of excessive coccinellin would generate toxicity against organisms.
