烟草柱头、花柱生理生化成分初步研究

摘    要:为研究烟草的花柱和柱头中生理成分在不同时期的变化情况,采用苯酚法、考马斯亮蓝法、茚三酮溶液染色法、福林法、愈创木酚法分别测定了4个烟草种不同发育时期柱头、花柱中的可溶性糖、蛋白质、氨基酸,酚类含量和POD同工酶活性。试验结果表明,不同烟草种间及同种不同发育时期各生理成分含量或活性具有显著差异,变化没有规律性。测定的所有烟草种中,所有种同时期柱头中氨基酸含量低于花柱,除N. stocktonii外,其他种同时期柱头上可溶性糖含量低于花柱。测定的所有种柱头上酚类含量显著高于花柱;除N. repanda外,其他种柱头中POD活性高于花柱,‘K326’及其雄性不育系同时期柱头中蛋白质含量显著高于花柱。

Abstract:The status and content of physiological components in the stigmas and styles were important to the germination and growth of pollen tubes. Soluble sugar, protein, acids, phenolic contents and POD isozyme activity in stigmas and styles of different Nicotiana species was determined by phenol method, coomassie brilliant blue, ninhydrin solution staining method, the forint method and guaiacol method. The results showed that the content and activity of different physiological components changed irregularly by the species and development stages. Acids content in stigmas was lower than that in styles among all the Nicotiana species and the same was true of soluble sugar apart from N. Stocktonii. On the contrary, phenolic contents in stigams was higher than that in styles and the same was true of POD isozyme activity apart from N. repanda. Protein contents in stigmas of [(gla.s) K326] and K326 was higher than that in styles.
