小白菜高效子叶离体植株再生体系的建立

摘    要:以丰顺小白菜带柄子叶为外植体,研究了苗龄、不同激素配比、A gNO3浓度和抗生素、共培养时间对不定芽再生的影响。结果显示,子叶不定芽再生的最佳苗龄为5~9d;经过2d的预培养,在M S 1.0m g/L TDZ 0.1m g/L NAA 7.5m g/L A gNO3 0.5m g/L ABA培养基中,带柄子叶不定芽再生率最高,为82.53%;在含有2.0m g/LIBA的1/2M S0培养基中诱导生根,不定根形成率为100%。抗生素敏感性试验表明,可选择浓度为20m g/L Hyg(潮霉素)作为最适合选择压,抑制农杆菌生长的抗生素选用500m g/L Cb(羧卞青霉素)。不同浓度农杆菌菌液对子叶再生不定芽的影响不大,但共培养时间的长短却明显地影响不定芽的分化。

Abstract:The cotyledons with petiole from Fengshun Chinese cabbage were used as explants,and the effects of seedling age and different homone combinations,AgNO_3 concentration and antibiotics,co-culture days with Agrobactium tumefaciens on the regeneration of adventitious bud of cotyledon were studied.The results indicated that the best seedling age to induce adventitious bud regeneration was 5-9 days,and the regeneration rate of cotyledon was the highest with 82.53% in medium MS 1.0mg/L TDZ 0.1mg/L NAA 7.5mg/L AgNO_3 0.5mg/L ABA after sub-cultured for two days;the formation rate of adventitious root was 100% in 1/2MS_0medium containing 2.0mg/L IBA for root induction.The results of antibiotics sensitivity experiments indicated that 20mg/L Hyg was the most appropriate for selective pressure and 500mg/L Cb was used to restrain the growth of Agrobactium tumefaciens.In addition,the differentiation of adventitious buds was evidently affected by co-culture days,but had no close relation with the concentration of Agrobactium tumefaciens.
