甜叶菊离体诱变与分子初检

摘    要:以甜叶菊茎段为外植体,用不同浓度的 NaN3对其进行不同时间的处理,筛选出合适的离体化学诱变体系,并对该体系下的部分再生苗进行RAPD和SRAP检测。结果表明:NaN3处理的半致死剂量为9 mmol/L浸泡4 h,该处理下植株矮化、腋芽数减少;用2种分子技术对该处理下14株再生苗的进行检测,结果表明,共28.6%再生苗的DNA序列发生了一定程度的变异,这为使用NaN3对甜叶菊进行大规模的离体诱变奠定技术基础。

Abstract:The stem of Stevia rebaudiana Bertoni in vitro as explants, different concentrations of NaN3 and treating time were used to optimize the mutation system. The results showed that the death rate about 50% appeared in stem sections treated with 9 mmol/L of NaN3 for 4 hours. The new plantlets became dwarf and less axillary buds appeared. And 14 plants tested by RAPD and SRAP indicated that the mutation rate reached 28.6%, which laid the foundation for mutation of Stevia rebaudiana Bertoni in vitro in large scale.
