黄瓜农杆菌介导法与花粉管通道法转基因技术

摘    要:采用农杆菌介导法和花粉管通道法进行抗虫基因EQKAM转入黄瓜的研究.优化了以子叶节为外植体的农杆菌介导转化技术;比较了切割柱头、子房注射和子房涂抹3种花粉管通道法处理授粉后黄瓜子房的转化效果,切割柱头和子房注射均可成功获得黄瓜抗虫转基因植株.PCR及斑点杂交证明EQKAM基因已整合到黄瓜基因组中.

Abstract:Transformation of insect resistant gene EQ KAM to cucumber was studied. Medium for A.tumefaciens mediated transformation using cotyledonary node as explants was optimized;3 methods including cutting chapiter,ovary injection and chapiter spreading for pollen tube pathway was compared. Cutting chapiter and ovary injection can both achieve insect resistant gene tranformants. PCR and dot blot test indicated that gene EQKAM had been incorporated into cucumber genome.
