摘    要:为了得到马铃薯抗病材料,本研究利用根癌农杆菌(Agrobacteriumtumefaciens)LBA4404转化马铃薯栽培品种“台湾红”,建立了优良的遗传转化系统:根癌农杆菌浓度以D600nm=0.5 ̄0.8为宜,感染时间以5min为宜;茎段和叶盘的预培养时间分别以4d和8d为宜,共培养时间皆以3d为宜。通过PCR和Sourhernblotting检测证明抗菌肽基因SPCEMA已成功导入其中。其茎段和叶片为的转化频率分别为10%和5.79%。

Abstract:In order to search an antibacterial potato (Solanum tuberosum L.), Various conditions for potato transformation with Agrobacterium tumefaciens LBA4404 were investigated and optimal conditions for the cultivar " Taiwanhong" were founded. At the same time, an efficient transformation system was developed. 1)The D600 of the Agrobacterium suspension was adjusted to 0.5~0.8 and then stems and leaf discs were dropped into the suspension for 5 min. 2)Stems and leaves were pre-cultured for 4 days and 8 days respectively. 3)The duration of co-culture is 3 days. Through this system SPCEMA gene was introduced into "Taiwanhong" and this was confirmed by PCR and Southern blotting. The transformation frequency of the stems and leaves was 10.00% and 5.79% respectively.
