| 摘 要： | 用曼氏血吸虫抗原特异性的保护性单抗Ｄ６多次免疫新西兰大白兔，从１只免疫兔分离得到的免疫血清在ＥＬＩＳＡ试验中对单抗Ｄ６的反应，比对血吸虫的其它两株同亚型抗原特异性单抗Ｎ（１２２）和Ｎ（３４）的反应强，能强烈地抑制单抗Ｄ６识别曼氏血吸虫尾蚴抗原（ＣＡＰ）和虫卵抗原（ＳＥＡ），而不能抑制单抗Ｎ（１２２）和Ｎ（３４）识别ＣＡＰ和ＳＥＡ。多次实验的结果还证明，单抗Ｄ６免疫兔血清不能识别ＣＡＰ和ＳＥＡ，正常兔血清不能抑制单抗Ｄ６识别ＣＡＰ和ＳＥＡ。单抗Ｄ６免疫兔血清经饱和硫酸铵和结合有正常小鼠血清蛋白的免疫亲和层析柱纯化后，其免疫原性不变。这些结果说明，免疫兔血清中含有抗保护性单抗Ｄ６的抗独特型抗体。日后可用它免疫动物，探索抗独特型抗体疫苗在血吸虫病免疫预防上的应用价值。

| Abstract: | Using the S. mansoni antigen specific MAb D 6 to immunize rabbits. we found that the serum from one immunized rabbit responded much stronger to the MAb D 6 than to the other two MAbs (N 122 and N 34) which have the same isotype with the MAb D 6 and are also specificto the S. mansoni antigen. The immunized rabbit serum also strongly inhibited MAb D 6 recognizing S. mansoni cercaria antigen preparation (CAP) and soluble egg antigen (SEA). but not to the MAbs N 122 and N 34. At the same time, we also noticed that the immunized rabbit sedid not respond to CAP and SEA. and the normal rabbit serum did not inhibit MAb D 6 recognning CAP and SEA. The results suggest that there are anti-D 6 idiotype antibodies in the Mab D 6 immunized rabbit serum. |
