家蚕糖转运蛋白BmST5基因的克隆与转录活性检测

摘    要:根据GenBank已登陆的糖转运蛋白序列与家蚕基因组框架图和EST序列,获得家蚕同源糖转运蛋白基因BmST5。基因编码区长1 527 bp,编码508个氨基酸,预测蛋白分子质量为55.67 kD。序列分析结果显示,该基因有典型的Sugar_tr结构域和12个疏水的跨膜结构域,与体虱等同源蛋白的一致性在50%以上。RT-PCR结果表明,该基因仅在家蚕的马氏管、生殖腺和头中有表达,在中肠、丝腺、血液、体壁和脂肪体中不表达。

Abstract:According to the reported sugar transporter sequences and silkworm scaffold and EST database,the gene homology of BmST5 in silkworm was obtained.The ORF is 1527bp in length and the putative encoding protein contains 508 AA and the MW is about 55.67kD.Sequenceanalysis showed that a typical Sugar_tr domain and 12 hydrophobic transmembrane domains exist in BmST5.Homology analysis of the deduced amino acids showed above 50% identity with Pediculus humanus corporis.The results of RT-PCR showed that the gene expressed in malpighian tubules,gonad,and head.No expression in other detected tissues,including midgut,silk glands,hemocyte,integument,and fat body was detected.
