灵芝多糖含量检测条件及其提取工艺的优化

摘    要:优化了苯酚-硫酸法测定灵芝多糖含量的显色条件,并且通过单因素和正交试验确定了灵芝多糖的最佳提取工艺条件。结果表明:苯酚-硫酸法测定灵芝多糖含量的条件为最大吸收波长490 nm,显色时间20 min,显色温度100℃;灵芝多糖的最佳提取工艺条件为提取温度80℃,提取时间2.5 h,液料比50 ml/g,在此条件下灵芝多糖的提取率为2.58%。

Abstract:The color condition for detecting polysaccharides in Ganoderma lucidum by phenol-sulferic acid method was optimized.The optimum extracting condition was obtained through single effect factor and orthogonal experiments.The condition of phenol-sulferic acid method was 490 nm maximum absorption wavelength,20 min reaction time and 100 ℃ reaction temperature.The optimum extracting condition was 50 minutes,60 ℃ and 50 ml/g liquid-material ratio.Extraction ratio of polysaccharides under the above condition was 2.58 %.
