| 摘 要： | 摘 要:从矮牵牛衰老花瓣中有效提取总RNA,是研究调控矮牵牛花瓣衰老相关基因逆转录PCR(RT-PCR)及其它分子生物学实验的前提.矮牵牛衰老花瓣富含色素等物质,提取时对传统的Trizol法进行了改良.利用普通琼脂糖凝胶电泳及紫外分光光度计对总RNA的完整性和浓度进行了检测.总RNA经电泳检测后,28S和18SrRNA条带清晰可见,且两者亮度比在1.0-2.0之间;总RNA用DEPC处理水稀释后,紫外吸光度D260/D280在1.75;经RT-PCR后,电泳检测,出现了清晰的目的条带.改良的Trizol试剂法能有效提取矮牵牛衰老花瓣RNA,且提取的总RNA可经RT-PCR成功的扩增出目的片段.

| Abstract: | In order to isolate the conservative fragments of ACC oxidase gene ofPetunia hybrida Vilm flower petals, to construct ACC oxidase gene RNAi expression vector ofPetunia hybrida Vilm.
