摘    要:本实验以猪带绦虫 TS11抗原基因的真核表达型质粒 VTS11肌肉免疫注射 BAL B/ c小鼠 ,四甲基偶氮唑蓝 (MTT)比色法检测小鼠脾淋巴细胞刀豆蛋白 A(Con A)刺激的增殖反应及 IL- 2的诱生活性 ;用 EL ISA方法检测免疫小鼠 Ig G总量和特异性抗体水平 ,常规法检测外周血免疫细胞数量的动态变化。结果显示 VTS11免疫小鼠血清的特异性抗体滴度和 Ig G含量显著高于空白对照组小鼠 ;免疫小鼠脾脏淋巴细胞 Con A刺激增殖反应和 IL - 2诱生活性均比对照组小鼠显著增强 ;VTS11免疫小鼠的淋巴细胞和巨噬细胞等免疫细胞的数量也显著超过对照组。这表明 VTS11免疫小鼠 ,可诱导其产生特异性的细胞和体液免疫反应 ,VTS11具有很强的免疫激活作用 ,可望成为预防猪囊虫病的一种新型疫苗。

Abstract:BALB/c mice at 2 months old were randomly divided into two groups and twice inoculated intramuscularly with different plasmids at the interval of 14 days. Group A was injected with VR1020, a eukaryotic expression plasmid, as control group. Group B was injected with VTS11, a recombinant plasmid containing a TS11 antigenetic gene of cysticercus cellulosae. Mice were bled on 0,7,14,21,28 and 35 days after immunization. The experimental results showed that the amounts of total IgG and specific antibody of the immunized mice were remarkably higher than those of the control group; the proliferative activity and the level of induced interleukin-2 bioactivity significantly increased in the lymphocytes of spleen from the immunized mice. Lymphocytes, macrophages and neutrophils of immunized mice have remarkably elevated in comparison with those of the control. These indicated that DNA vaccination with VTS11 significantly enhanced the humoral and cellular immune responses of mice,So TS11 gene could be employed as a hopeful immunoprotective gene in the further development of DNA vaccine against swine cysticerosis.
