摘    要:通过探讨pH值对甜橙花粉萌发和花粉管生长及其钙离子分布的影响,为进一步研究甜橙授粉受精机制提供依据,为甜橙人工授粉等生产实践提供指导。试验以4个甜橙品种花粉为试材,对外源pH值为4.0、5.0、6.0、7.0、8.0、9.0时花粉萌发率和花粉管平均生长速率进行观察统计并测定其花粉管尖端钙离子分布及其相对荧光强度值。结果表明,甜橙花粉能在外源pH值为4.0~9.0的广泛条件下萌发;品种434在pH 7.0时花粉萌发率最高;其他3个品种则在外源pH 6.0时最高;各品种在pH 6.0~7.0的范围内有较高的钙离子相对荧光强度并保持花粉管尖端游离钙离子浓度的梯度分布,从而保证后期花粉管的持续生长。

Abstract:The paper discussed the influence of the pH to the orange pollen germination and pollen tube growth and the distribution of Ca2+ aiming to provide some basis for the further study on orange pollination fertilization mechanism and to offer some help for orange artificial pollination and other production practice.Using four kinds of orange pollens as material,the pollen germination rate and pollen tube growth speed at different extracellular pH level of 4.0,5.0,6.0,7.0,8.0 and 9.0 was observed and computed.The calcium distribution in the tip of pollen tube and its relative fluorescence intensity was measured.The results showed that orange pollen could germinate under the exogenous pH of 4.0~9.0.Cultivar 434 had the best germination rate with pH 7.0,and the other cultivars had best germination rate under pH 6.0.Four cultivars had higher relative Ca2+ fluorescence intensity and kept the Ca2+ concentration in gradient distribution,thus can ensure the sustainable growth of late pollen tubes.
