牡丹AFLP荧光反应体系的建立和优化

摘    要:以牡丹的叶片为材料,通过对扩增长度多态性(AFLP)反应体系中几个关键参数进行优化,建立了牡丹的AFLP荧光反应体系,首次在牡丹DNA提取、酶切连接、预扩增、选择性扩增等试验过程取的成功,得到了清晰的部分牡丹品种的指纹图谱.为牡丹品种指纹图谱的绘制、亲缘关系的研究等奠定基础.

Abstract:The improved method of CTAB-DNA isolation was used to extract total DNA from leaves of Paeonia suffruticosa in this study.AFLP analysis in Paeonia suffruticosa was established.Clear and high resoluteion DNA fingerprints were obtained in this study,which made a success in DNA extraction,restriction-ligase reaction,pre-amplification and staining techniques.The aim of this paper was to lay a foundation for the establishment of DNA fingerprinting,selection and breeding of eminent cultivars and genetic relationship in Paeonia suffruticosa.
