金银忍冬茎和叶中绿原酸的提取及含量测定

摘    要:[目的]为寻找绿原酸新药源、综合利用金银忍冬提供科学依据。[方法]采用超声技术提取金银忍冬茎、叶中的绿原酸,高效液相色谱法对绿原酸含量进行测定。[结果]检测波长选择为327 nm。绿原酸进样量与峰面积在0.20~1.00μg范围内具有良好的线性关系。精密度试验的RSD为1.30%,说明仪器较精密。稳定性试验的RSD为1.15%。加样回收率试验的RSD为1.35%,平均回收率为98.52%。金银忍冬叶和茎中均含有绿原酸,经外标法测定其含量依次为:叶(1.54%)>茎韧皮部(0.44%)>茎木质部(0.42%)。[结论]超声提取法简单、快速,检测方法可行,测定误差较小,结果较准确,重现性好,可用于金银忍冬茎和叶中绿原酸的定量分析。

Abstract:[Objective] The objective was to provide a scientific basis for the search of new resource of chlorogenic acid and the multiple utilization for Lonicera maackii(Rupr.) Maxim.[Method] Chlorogenic acid was extracted from the stems and leaves by ultrasonic technology and its content was determined by HPLC.[Result] The detecting wavelength was selected as 327 nm,the sample volume of chlorogenic acid had good linear correlation with the peak area within 0.20-1.00 μg.RSD value of precision experiment was 1.30%,and it indicated that the instrument was precise.RSD value of stability test was 1.15% and that of spotting recovery test was 1.35%,with the average recovery rate of 98.52%.The stems and leaves of L.M.Maxim all had chlorogenic acid with the content from big to small being leaves(1.54%) >stem phloem(0.44%)>stem xylem(0.42%),which was determined by external standard method.[Conclusion] Ultrasonic technology was a simple, rapid and feasible method with small determinate error,accurate result and good reproducibility,and could be applied to the quantitative analysis of chlorogenic acid in the stems and leaves of L.M.Maxim.
