花生幼苗多胺氧化酶的分离提取

摘    要:对花生(Arachis hypogaeaL.)幼苗多胺氧化酶(Polyamine oxidase,PAO)的分离提取方法进行了研究,内容涉及PAO活性在花生幼苗各器官中的分布以及在不同萌发时期的变化、酶提取液组分对PAO提取效果的影响.结果表明,在黑暗条件下萌发的花生幼苗,其胚根和胚轴的PAO活性分别在萌发后的第4 d和第5 d出现一个峰值;含1%PVP和0.5 M NaCl的磷酸缓冲液(0.1 mol/L,pH7.0)用作花生PAO分离提取液可取得较好效果.

Abstract:A series of experiments concerning the extraction of polyamine oxidase from peanut(Arachis hypogaea L.) were conducted,including distribution of PAO activity in peanut young seedlings and its change during the germination period.It was found that the highest PAO activities in embryonic axes and radicles of the peanut seedlings occurred in day 4 and day 5 respectively after germination in dark.Additionally,the extraction medium of peanut PAO was 0.1 mol/L phosphate-buffered saline(pH7.0),containing 0.5 mol/L NaCl and 1%(g/V) polyvinylpyrrolidone.
