摘 要: | 以溪荪为试验材料,研究了消毒时间、外植体种类对溪荪组织培养的影响,筛选出了适宜的初代培养基、增殖培养基和生根培养基。试验结果表明:适宜的外植体为带芽根状茎,以1g/L的HgCl2消毒8min效果最好。适宜的初代培养基为MS+BA1.0mg/L+NAA0.2mg/L,诱导成苗率达88.57%。适宜的增殖培养基为MS+BA2.0mg/L+NAA0.2mg/L,增殖系数为4.72。适宜的生根培养基为MS+BA0.1mg/L+NAA0.5mg/L,平均每株生根数量为8.9条,平均根长为2.5cm,平均根粗为1.0mm。

Abstract: | Taking Iris sanguinea as experimental material, effects of disinfection time and explants type on tissue culture were studied, and the optimum initial medium.multiplication medium and rooting medium were also selected. Results indicated that rhizome with buds was the optimum explants, and the optimum disinfection time with lg/L HgC12 was 8 rain. The optimum initial medium was MS~BA1.0mg/Lq-NAA0.2mg/L, and the induction rate was 88.57%. MS+BA2.0mg/L+NAA 0.2mg/L was suitable for multiplication, with the multiplication coefficient of 4.72. The regenerated plantlets rooted well in MS-kBA0. lmg/L+NAA0. 5mg/L, the average number of root was 8.9, the average length and diameter of root was respectively 2.5cm and 1.0ram. |
