非洲堇无根苗的生根方法研究

摘    要:目的]建立非洲堇无根苗的生根技术体系。方法]以非洲堇的无根苗为试材,研究组培法、水培法、扦插法和滤纸桥法4种生根方法对非洲堇生根的影响。结果]4种生根方法均能诱导非洲堇无根苗生根,水培法产生不定根的平均根长(2.5cm)和平均生根数(42条)均显著高于其他方法。扦插法产生的不定根直径大、长势旺,移栽后成活率高达99.3%,可以应用于大规模生产。40ml营养液的生根数量最大,但植株高度和根的长势均劣于30ml营养液无根苗,应选择适宜的营养液体积。结论]以珍珠岩为基质在试管外扦插非洲堇生根是可行的,具有广阔的应用前景,对于降低非洲堇大批量生产的成本有重要意义。

Abstract:Objective The aim of the research was to set up rooting technology system of the rootless seedlings of Saintpaulia ionantha.Method With the rootless seedlings of S.ionantha as test materials,the effects of 4 kinds of rooting methods as tissue culture method,water culture method,cutting method and filter paper bridge culture method on the rooting of S.ionantha were studied.Result 4 kinds of rooting methods could all induce the rooting of the rootless seedlings in S.ionantha and average root length(2.5 cm)and average root number(42 pieces)of adventitious root produced by water culture method were higher than that of other methods.The adventitious root produced by cutting method had great diameter and strong growth vigor and its survival rate after transplanting reached 99.3 %,which could be used in mass production.The rooting quantity of rootless seedlings in 40 ml nutrient solution was most,but its plant height and root growth vigor were more inferior than that of the rootless seedlings in 30 ml nutrient solution.Therefore,the appropriate volume of nutrient solution should be chosen.Conclusion With perlite as medium,the rooting of S.ionantha by using in vitro cutting was feasible and had broad application foreground,which had great significance to reduce the mass-production cost of S.ionantha.
