欧洲卫矛愈伤组织分化及其生理生化特性

摘    要:以欧洲卫矛生长健壮的叶片愈伤组织为材料,通过不同激素配比筛选出适宜其分化的培养基,将愈伤组织置于不同条件下培养。结果表明,适宜欧洲卫矛愈伤组织分化的培养基为MS+1.0 mg·L-1 6-BA+0.1 mg·L-1 NAA;欧洲卫矛愈伤组织适宜在光照条件下进行培养,生长周期为28 d,28 d后愈伤组织出现不同程度的褐化;可溶性糖含量先升后降;SOD和POD活性均出现2个峰值,可溶性蛋白变化趋势与POD变化趋势基本一致;可溶性蛋白在分化中起重要作用,且光照条件下,可溶性糖和可溶性蛋白含量、SOD和POD活性均高于黑暗条件下,PPO在培养后期活性下降,可能与愈伤组织褐化有一定关系。

Abstract:In the study, a suitable differentiation medium was obtained, using the callus of the healthy leaves of Euonymus europaea cultured under different conditions with different hormone ratios. The results showed that: the suitable medium for callus differentiation of Euonymus europaea was MS+1.0 mg·L-1 6-BA+0.1 mg·L-1 NAA; the callus of Euonymus europaea was suitable for culture under light with growth cycle of 28 days, and it would appear browning in different degrees after 28 days; the soluble sugar content in the callus rose firstly and then decreased; the activities of SOD and POD in the callus both showed two peaks, and the change trend of soluble protein was basically consistent with the POD activity; soluble protein played an important role in the callus differentiation, and the contents of soluble sugar and soluble protein, the activities of SOD and POD were higher grown under illumination than those under dark conditions. The activity of PPO decreased in the late stage of culture, which may be related to the browning of callus.
