摘    要:相关序列扩增多态性(sequence—related amplified polymorphism,SRAP)是基于基因的内含子和外显子区域进行检测的新型分子标记技术,具有多态性高、重复性好、便于克隆目标片段等特点,鉴于甜瓜(Cucumis melo L.)中还鲜有SRAP-PCR的相关研究报道。本研究利用L16(4^5)正交设计对甜瓜SRAP-PCR反应体系进行了优化。结果确定了10μLSRAP-PCR的最优反应体系:200ng/μL模板DNA2.5μL、10μmol/L引物0.3μL、5U/μL Toq DNA聚合酶0.6μ20mmol/L Mg^2+ 1μL和2mmol/L dNTP 2μL。这一优化的反应体系为今后利用SRAP-PCR技术对甜瓜进行相关研究提供了帮助。
