黑枸杞花青素研究进展

摘    要:目的研究青藏高原黑枸杞花青素对小鼠巨噬细胞RAW264.7的凋亡作用及相关分子机制。方法利用扫描电镜、DAPI和Hoechst染色、流式细胞术、RT-qPCR和Western-blot检测不同质量浓度黑枸杞花青素诱导小鼠巨噬细胞24 h后对细胞形态学、细胞凋亡率、JNK信号通路因子及相关因子mRNA和蛋白表达的影响。结果花青素诱导细胞24 h后未见明显凋亡形态,且细胞数量增多,细胞凋亡率极显著下降(P<0.01);随着黑枸杞花青素质量浓度增加,Caspase-3 mRNA及蛋白表达降低,Bax/Bcl-2 mRNA表达量比值降低,JNK通路因子JNK mRNA及p-JNK蛋白表达降低且呈剂量依赖性;JNK抑制剂组能显著降低JNK通路因子JNK mRNA (P<0.05)及p-JNK蛋白表达(P<0.01);与JNK抑制剂组相比,花青素+SP600125组能显著降低JNK通路因子JNK mRNA (P<0.05)及p-JNK蛋白表达(P<0.01)。结论黑枸杞花青素可能通过JNK通路抑制小鼠巨噬细胞RAW264.7凋亡从而发挥免疫调节作用。

Abstract:PurposeTo study the effect of anthocyanins from Qinghai-Tibet Plateau black wolfberry on the apoptosis of mouse macrophages RAW264.7 and related molecular mechanisms. MethodScanning electron microscopy, DAPI and Hoechst staining, flow cytometry, RT-qPCR, and Western-blot were used to detect the effects of different mass concentrations of black wolfberry anthocyanins on the cell morphology, apoptosis rate, the influence of mRNA and protein expression of JNK signaling pathway and related factors, after 24 hours induction on mouse macrophage. ResultThere was no obvious apoptotic morphology after 24 hours of anthocyanin-induced cells, and the number of cells increased, and the apoptosis rate was extremely significantly reduced (P<0.01). As the mass concentration of black wolfberry anthocyanins increased, Caspase-3 mRNA and the protein expression decreased, the ratio of Bax/Bcl-2 mRNA expression decreased, and the mRNA and p-JNK protein expression of JNK pathway factor JNK decreased in a dose-dependent manner. The JNK inhibitor group could significantly reduce the mRNA (P<0.05) and p-JNK protein expression of JNK pathway factor JNK (P<0.01); compared with the JNK inhibitor group, the anthocyanin+SP600125 group could significantly reduce the mRNA (P<0.05) and p-JNK protein expression of the JNK pathway factor JNK (P<0.01). ConclusionThe black wolfberry anthocyanin maybe inhibit the apoptosis of mouse macrophages RAW264.7 through the JNK pathway to play an immunomodulatory effect.
