摘    要:新疆地域广阔,各地气候条件差异大.西瓜、甜瓜在新疆种植面积较广,其病毒病的发生和危害较重.西瓜花叶病毒2号(Watermelon mosaic virus 2,WMV-2)是致病的主要毒源之一.前人对WMV-2的研究仅限于病原鉴定以及它对瓜类氨基酸的影响,而对它的遗传变异未做研究.为了深入研究新疆不同地域WMV-2致病性的差异,有必要弄清楚新疆的WMV-2有无遗传变异的发生.PCR-SSCP是一种基于PCR的单链构象多态性(single-stranded conformation polymorphism)分析技术,主要用于DNA已知突变的检测或未知变异的分析1].

Abstract:Twelve isolates of Watermelon mosaic virus 2(WMV-2) were collected from Cucurbitaceae plants in different areas of Xinjiang Uygur Autonomous Region in 2008, and their genetic variations were analyzed by RT-PCR-SSCP. A specific fragment of about 421 bp was amplified among all the isolates by RT-PCR. Using analysis of single-strand conformation polymorphism (SSCP), 12 isolates of WMV-2 were classified as five variant types: KS2 ( typeⅠ), S7( type Ⅱ), S4( type Ⅲ), KS1 and S6 (type Ⅳ), M1,M2,K1,S3,S5,S1 and S2(type Ⅴ). It was shown that there were genetic variations and differences in regional distribution among the isolates of Cucurbitaceae WMV-2 in Xinjiang, and there was obvious differentiation among the isolates of the different Cucurbitaceae plants in the same place.
