摘    要:以新台糖22的胚性愈伤组织为材料,在诱导培养基、分化培养基及生根培养基中,分别添加不同浓度的潮霉素(Hyg)为选择性试剂。以确定甘蔗遗传转化时的最佳筛选浓度。结果表明:潮霉素对不同阶段培养基的敏感性存在一定差异,在甘蔗遗传转化时3种培养基的抗性筛选浓度分别以50、30、30mg/L为佳;在分化培养基内的毒性作用比在诱导培养基内更快、效果更好,故遗传转化时可直接将Hyg用于分化培养基进行抗性芽筛选试验。

Selective Test of Hygromycin B in Different Stages of Sugarcane Genetic Transformation

Abstract:In order to get adaptive concentrations of selective reagent, different concentrations of hygromycin B (Hyg) were added to each medium of sugarcane tissue culture with embryonic callus of sugarcane variety ROC22 as the experimental material. The results showed as follows: among different medium of sugarcane genetic transformation, there were certain differences existed to Hyg sensitivity, the ideal selecting concentrations of Hyg were 50 mg/L differentiation in callus growth, 30mg/L in bud differentiation and plantlet was more susceptible to Hyg than that of callus growth, so we rooting, respecting. The stage of bud should select medium differentiation in study of selective test.
