摘    要:以铁皮石斛类原球茎为材料,研究了在培养过程中添加不同质量浓度的蔗糖对铁皮石斛原球茎干物质量、多糖积累动态变化过程及原球茎在改变渗透压的条件下抗氧化物酶变化的影响,以期找出在培养过程中添加的最适蔗糖质量浓度,以提高原球茎的多糖产量。结果表明:在培养第20天添加5 g/L蔗糖对原球茎多糖积累最为有利,其干质量及多糖含量、多糖产量最高,分别为16.02 g/L、300 mg/g、4 805.85 mg/L。同时,在培养第20天添加5 g/L蔗糖对铁皮石斛原球茎抗性的提高最为有利。

Abstract:The effects of different sucrose concentration feed on the dry weight,the accumulation of polysaccharides and the activity of antioxidant enzyme with different osmotic pressure during the culture process were studied in this experiment of which protocormn-like bodies of D.candidum was used as material.The purpose of the experiment is to find the most appropriate concentration of sucrose in the process of culture in order to increase polysaccharide production of protocormn-like bodies of D.candidum.The results showed that intermittent feed 5g/L sucrose after 20 days culturing was benefited to the accumulation of polysaccharides of protocormn-like bodies of D.candidum.The dry weight,contents of polysaccharides and production of polysaccharides reached to it's maximum with 16.02g/L,300mg/g,4805.85mg/L respectively.Meanwhile intermittent feed 5g/L sucrose after 20 days culturing was benefited to increase the resistance of protocormn-like bodies of D.candidum.
