大豆根系分泌物的鉴定及PAL1,PAL2,C4H的克隆

摘    要:为研究大豆根系分泌物在大豆连作障碍中的作用,利用土壤溶液原位取样器收集已灭菌的大豆正茬、一年重茬和两年重茬土壤盆栽的大豆根系分泌物,并采用GC-MS进行定性分析,检测出正茬、一年重茬和两年重茬的大豆根系分泌物,分别有355、7和64种,均含有烃、醇、酸、酯、酚、苯和醛等化合物。将收集的正茬、一年重茬和两年重茬的大豆根系分泌物溶液浇灌大豆幼苗,发现对茎长、茎鲜重、根长、根鲜重、光合速率和叶绿素含量均有一定的抑制作用。同时,以二氯甲烷(CH2Cl2)为溶剂,提取培养28d的大豆根系分泌物,共分离鉴定出大豆根系分泌物77种,其中包括烷烃、醇、酸、酯、苯、酚、萘、酰胺、酮、醛类以及其他类化合物。其中酚类物质占0.03%。基于追溯苯酚和苯甲酸类物质代谢的关键酶,克隆了苯丙氨酸解氨酶PAL家族的两个成员PAL1和PAL2,以及苯丙氨酸代谢的第二个关键酶C4H基因,为后续的基因操作提供依据。
