海洋多粘类芽孢杆菌L1-9抗菌活性产物的分离与结构鉴定

摘    要: 为了从海洋多粘类芽孢杆菌发酵液中分离鉴定抗菌活性产物,以对禾谷镰刀菌(Fusarium graminearum)的生物活性进行跟踪,采用硅胶柱层析、Sephadex LH-20柱层析、薄层层析和HPLC等技术,对海洋多粘类芽孢杆菌(Paenibacillus polymyxa)L1-9抗菌活性组分进行分离纯化,采用ESI-MS、1H NMR、13C NMR等波谱分析技术,对活性组分进行结构鉴定。采用牛津杯法测定活性组分的抑菌谱。得到了2种具有强抗菌作用的组分B1和B2;认为组分B1和B2分别为邻苯二甲酸二丁酯(DBP)和邻苯二甲酸二(2-乙基己基)酯(DEHP)。DBP和DEHP对供试的5种病原真菌和细菌具有抑制作用。邻苯二甲酸二丁酯的抑制作用高于邻苯二甲酸二(2-乙基己基)酯。

Abstract:The antibacterial metabolites from fermentation broth of marine Paenibacillus polymyxa strain L1-9 were purified by silica gel column chromatography, Sephadex LH-20, thin layer chromatography and HPLC methods, and the structures of these metabolites were identified by ESI-MS, 1H NMR、13C NMR and spectra analysis. The antibacterial activities were then determined by Oxford cup method. It was demonstrated that two kinds of compounds, B1 and B2 showed strong antibacterial activities, which were identified as dibutyl phthalate (DBP) and di(2-ethylhexyl)phthalate(DEHP), respectively. DBP and DEHP had inhibition effects on five kinds of fungi and bacteria. The inhibition effect of DBP was higher than that of DEHP.
