六月酥梨叶片培养和植株再生

摘    要:以六月酥梨叶片为外植体,研究了基本培养基、不同质量浓度BA和IBA的组合、不同暗培养时间、不同碳源种类和质量浓度、叶片放置方式和乙烯抑制剂AgNO3处理等因素对六月酥梨叶片再生体系的影响。结果表明,六月酥梨叶片再生芽的适宜组合为NN69+BA 5.0 mg/L+IBA 0.3 mg/L+山梨醇40 g/L+琼脂7 g/L,远轴面向下,暗培养30 d。在适宜条件下,六月酥梨叶片再生频率高达83.3%,平均再生芽数为2.84。不同碳源种类和质量浓度对六月酥梨叶片再生有不同的影响,其中40 g/L山梨醇对叶片不定芽诱导效果显著。硝酸银质量浓度为0.1~1.0 mg/L时六月酥梨叶片再生频率高于对照,但无显著差异;当硝酸银质量浓度为2.0 mg/L时,显著降低了叶片再生频率。叶片再生苗在生根培养基1/2MS+IBA 0.5 mg/L+蔗糖20 g/L+琼脂6.5 g/L,暗培养5 d,生根率为50%,平均生根数为3.33。

Abstract:Using the isolated leaves of Liuyuesu pear as the explants,plant regeneration from leaves of Liuyuesu pear in vitro were conducted to examine the factors,including medium type,concentration of plant growth regulators,carbon sources,dark culture period,ways of leaves inoculation and treatment of AgNO3 that influence adventitious buds regeneration from leaf.The results showed that the optimal combination for leaf regeneration of pear cultivar Liuyuesu was NN69+BA 5.0 mg/L+IBA 0.3 mg/L + sobitol 40 g/L + agar ...
