中华蜜蜂mrjp3基因cDNA的克隆及序列分析

摘    要: 以东方蜜蜂(Apis cerana)mrjp3基因部分片段为探针筛选中华蜜蜂(Apis cerana cerana,简称中蜂)8日龄工蜂头部cDNA文库,得到120个阳性克隆。对这些阳性克隆进行进一步的筛选和序列测定,获得含有中蜂MRJP3 (AccMRJP3)cDNA的阳性克隆,对阳性克隆的插入片段进行测序,得到AccMRJP3的cDNA序列全长。中蜂MRJP3的cDNA全长为1 987 bp(包括10 bp长的poly A尾巴),包含一个长1 779 bp,编码593个氨基酸的ORF,在5'端和3'端各有一个长46 bp和160 bp的非编码区。类似于西方蜜蜂(Apis mellifera)和大蜜蜂(Apis dorsata),由AccMRJP3 cDNA编码的氨基酸序列也存在一个长片段重复区。但比较结果显示中蜂、西方蜜蜂、大蜜蜂MRJP3重复区之间存在一定差异。

Abstract:By screening the worker(Apis cerana cerana)heads cDNA library, using a fragment of the mrjp3 gene of Apis cerana as probe, 120 positive clones were obtained. The clone containing AccMRJP3 cDNA was selected. Based on the sequencing of the inserts of the positive clone, a sequence of AccMRJP3 cDNA which is 1 887 bp long including a poly(A) tail was obtained. The AccMRJP3 cDNA encompassed an open-reading frame (ORF) with 1 779 bp encoding 593 amino acids. The un-translated regions (UTR) of the 5′end and 3′end are 46 bp and 160 bp long, respectively. Similar to AmMRJP3 and AdMRJP3, the putative AccMRJP3 also has a repetitive region. The comparison of the repetitive region of AccMRJP3, AmMRJP3 and AdMRJP3 shows some differences between them.
