摘 要: | [目的]探讨菲律宾蛤仔酶解多肽的制备及体外抗肺癌H1299细胞的活性。[方法]选择胰蛋白酶酶解菲律宾蛤仔,经超滤得到3kD以下的酶解多肽,应用MTT法、HE染色和AO/EB染色等方法检测其体外抗H1299细胞的活性。[结果]3 kD以下的酶解多肽作用于H1299细胞后,MTT法检测结果表明该多肽能抑制该细胞增殖,呈剂量和时间依赖;经HE染色和AO/EB染色发现H1299细胞出现凋亡的形态学改变。[结论]菲律宾蛤仔酶解多肽能明显抑制H1299细胞的增殖并诱导细胞凋亡。

Abstract: | [ Objective ] To discuss the preparation of peptide from Ruditapes philippinarum and anti-lung cancer H1299 cells activity in vitro. [ Method] Trypsin was used to enzymolysis, 3 kD molecular weight cut-off membranes were used to fractionate the Ruditapes philippinarum hy- drolysate, and 〈 3 kD weight fractions were obtained. The anti-proliferation activity and the early apoptosis of Ruditapes philippinarum peptides were detected by MTF, HE staining and AO/EB double staining, respectively. [ Result] MTF assay showed that the peptide inhibited prolifera- tion of H1299 cells in both concentration-dependent and time-dependent manners; Cell apoptosis morphological feature was observed under HE staining and AO/EB double staining, and staining showed that the peptide induced cellular morphological change. [ Conclusion ] The Ruditapes philippinarum hydrolysate inhibited the lung cancer H1299 obviously and induction of apoptosis. |
