抗兔病毒性出血症病毒单克隆抗体的制备

摘    要:通过抗兔病毒性出血症病毒(RHDV)杂交瘤细胞株的筛选,其中一株连续克隆3次,在培养瓶中传代1个月,用酶联免疫吸附试验(ELISA)测定,分泌单克隆抗体(McAb)性能稳定。杂交瘤细胞株的小鼠腹水滴度为1:1600~1:3200。利用McAb腹水作一抗,经免疫荧光检测RHDV感染的细胞片,可发现细胞核内有特异性荧光颗粒,滴度为1:200。杂交瘤细胞株染色体的分析,染色体数在98±10,符合杂交瘤细胞株的特征。经琼脂扩散法对McAb亚类的分析测定结果与抗鼠IgG和IgG_3出现沉淀线。

PREPARATION OF RAHDV MONOCLONIC ANTIBODY
Abstract:Hybridoma cell strains of rabbit antihaemorrhagic disease virus (RAHDV) have been screened. Of which, one was cloned continuously for three times and propagated over one month. The function of its firmly secreting McAb was determined by ELISA. Monoclonic antibody (McAb) is prepared from ascites of mice whose titer is 1:1600-1:3200. In nuclei of cells infected by RAHDV are found specific fluorescence particles, the titer is 1: 200. The chromosome number of hybridoma cell strain accounts for 98卤10. Immunodiffusion suggests that McAb makes precipitation reaction with IgG and IgG3.
