黄喉拟水龟摩氏摩根菌的分离鉴定及系统发育分析

摘    要:从广西某黄喉拟水龟(Mauremys muticaCantor)养殖场发病的黄喉拟水龟的肝、肺分离到一致病性的菌株(HX081027),经人工感染实验证实其为该病的病原菌。对该菌的形态、生理生化进行分析,试验结果与报道的摩氏摩根菌一致,表现为革兰氏阴性、两端钝圆的短杆菌,无荚膜及芽孢,葡萄糖产酸产气,接触酶、脲酶、苯丙氨酸脱氨酶和鸟氨酸脱羧酶阳性,氧化酶、脂酶、DNA酶和赖氨酸脱羧酶阴性,甲基红阳性,VP反应阴性,MR阳性等特征。采用扩增细菌16S rDNA的通用引物对该菌的16S rDNA基因进行PCR扩增、克隆测序,得到1条长度为1506bp的核苷酸序列(GenBank登录号为FJ858185)。以该菌16S rDNA序列和GenBank数据库内同源性较高的细菌16S rDNA序列进行同源性比对分析和构建系统发育树,结果显示分离菌HX081027与摩氏摩根菌属内相关菌株的同源性在98.8%~99.6%,且在系统发育树上与来源于鱼类的M.morganiiJCM1672聚为一支,结合形态及生理生化特点将其确定为摩氏摩根菌(Morganella morganii)。

Abstract:A pathogenic bacterium(HX081027)was isolated from the liver and lung of the diseased Mauremys mutica Cantor,and the bacterium was proved to be the pathogen of the disease through artificial infection.Its morphological,physiological and biochemical characteristics were studied,and they completely matched to all of the reference Morganella morganii(M.morganii)strain which has been reported.The bacteria are gram negative,rods,non-capsule and spore;Glucose-positive,catalase-positive,urease-positive,phenylalanin...
