皖西白鹅Pit-1基因插入/缺失多态性及其对早期体重的影响

摘    要:本文根据鸡Pit-1基因mRNA 序列设计一对引物,以皖西白鹅DNA为模板,扩增鹅Pit-1基因的部分片段,结果发现该片段存在2个插入/缺失突变,共出现3种基因型,分别为AA、AB和BB,与AA型相比BB型分别在扩增片段的132和148bp后插入3 bp和13 bp。卡方检验结果表明,皖西白鹅的基因型分布符合哈代温伯格平衡。不同基因型之间各期体重比较的结果表明,AB型、BB型个体3-11周龄体重均显著高于AA型(P<0.05);BB型个体3-11周龄体重高于AB型,但差异不显著(P>0.05),表明就影响鹅早期体重而言,B等位基因为有利基因。

Abstract:Goose Pit-1 gene was amplified in this study; the primer was designed according to the sequence of chicken Pit-1 mRNA. Two insert/deletion mutations were found in this fragment, and 3 genotypes were detected within Wanxi White goose, which were AA, AB and BB. Compared with AA genotype, there were 3bp and 13bp insertion after the position of 132bp and 148bp respectively in BB genotype. Deviation of Wanxi White goose from Hardy-Weinberg equilibrium was tested, and it was not significant (P>0.05). Comparison of bodyweight between different genotypes showed that bodyweight of 3-11 weeks in AB and BB genotype were significantly higher than AA(P<0.05), also BB genotype showed higher bodyweight than AB, but the difference was not significant (P>0.05), which suggest that B was the positive allele as for the effects on goose bodyweight.
