家蚕孤雌生殖系与有性生殖系亲本的血液比较蛋白质组学研究

摘    要:为进一步解析家蚕孤雌生殖的发生机制,采用双向电泳(2-DE)、基质辅助质量飞行时间质谱(MALDI-TOF/MS)和生物信息学技术,对高发生率和高孵化率的家蚕孤雌生殖系与其有性生殖系亲本的血液蛋白质组成进行比较分析。较其有性生殖系亲本,孤雌生殖系幼虫在5龄第3天、第5天和第7天血液中的差异蛋白不完全相同。对6个持续表达和差异明显的蛋白进行质谱分析,成功鉴定其中2个蛋白分别是保幼激素结合蛋白和胰凝乳蛋白酶抑制剂-8A,推测这2个蛋白及其余4个未被鉴定出的蛋白与2种生殖方式家蚕品系的生长发育、生殖等过程的调节有关联。

Abstract:To gain an insight into the mechanism of silkworm parthenogenesis,the hemolymph proteins of Bombyx mori parthenOgenetic line which had high incidence rate and hatchability were compared with its amphigenetic parent by pro- teomics approach including two dimensional electrophoresis(2-DE),matrix assisted laser desorption ionization and time of flight mass spectrometry(MALDI-TOF/MS),and bioinformatics analysis.The results showed that the differential pro-teins in hemolymph of the 5th instar larvae at day 3,5 and 7 of the parthenogenetic line were not identical with those of its amphigenetic parent.6 differential proteins which were expressed continually and had significant differences were aria-lyzed with mass spectrometry.Among them,2 proteins were identified successfully.They were juvenile hormone binding protein and chymotrypisn inhibitor-8A.It could be speculated that these 2 proteins and the remaining 4 non-identified proteins might be associated with the regulation of growth,development and reproductive processes in different repro- ductive modes of the silkworm,Bombyx mori.
