摘    要:谷子幼穗诱导形成的愈伤组织,经甲基磺酸乙脂(EMS)诱变处理和8个月的连续筛选及近6个月的稳定培养后,获得了一个耐盐性稳定的细胞变异系,并且获得了它的再生植株。对在无盐压下生长三代的耐盐细胞系的生理生化分析表明,其对氯化钠和聚乙二醇所引起的离子和水分胁迫具有较强的抗性。在无盐胁迫条件下,耐盐系的相对细胞

Abstract:A stable NaCl-tolerant cell line was obtained by treating the embryogenic calli ini-tiated from young inflorescence segments of millet (Setaria italica, Var.7520) with ethyl methane sulphonate (EMS) after screening for 8 months and stabilizing for about 6 months. The regenerated plants were produced from it. Physiological and biochemical analysis showed that, the NaCl-tolerant cell line grown on the NaCl-free medium for 3 generations exhibited stronger resistance to NaCl stress and water stress caused by PEG. When grown on NaCl-frce medium, the relative osmotic potential of tolerant cell line was lower than that of control cell line, however, free proline content and the activity of superoxide dismutase (SOD) were 105% and 90% higher than those in controls. The results of SDS electrophoresis of soluble proteins indicated that the amount of scanning bands of proteins in NaCl-tolerant cell line was more than that in control. New protein bands of 59kd and 90kd were observed in salt-tolerant cell line.
