摘    要:[目的]筛选适合作为TAIL-PCR模板的水稻基因组DNA提取方法。[方法]采用5种方法提取水稻叶片基因组DNA,筛选出DNA纯度及浓度较高的、适合作为TAIL-PCR模板的水稻基因组DNA提取方法。[结果]改良的SDS法及CTAB法提取的基因组DNA纯度及浓度较高,效果明显优于脲法及简易法。以TAIL-PCR技术对选择的同一个含Ds元件的样品的5种提取方法的基因组DNA作为模板进行Ds侧翼序列的扩增,改良的SDS法及CTAB法得到的条带特异,这些产物经回收、纯化后可直接用于序列的测定。但考虑到CTAB价格比较昂贵,毒性较大,一般不主张采用。[结论]改良的SDS法提取的水稻基因组DNA最适作为TAIL-PCR的模板。
