草菇开伞相关基因反义表达载体构建

摘    要:以质粒pCAMBIA1301和pBI121为基础构建草菇表达载体pCB。把已克隆的草菇开伞相关基因DNA片段正反向插入pCB,获得pCB—opn15和pCB—opn39,经测序证实,pCB—opn15为正向插入,pCB—opn39为反向插入。

Abstract: An expression vector of pCB for Volvariella volvacea was constructed with plasmid pCAMBIA1301and pBI121. The DNA fragment of fruitbody opening related gene (opn) was inserted to plasmid pCB by two orientation. Two recombined plasmids of pCB-opn 15 and pCB-opn 39 were obtained,and the sequencing results showed that pCB-opn 15 was inserted in positive orientation and pCB-opn 39 was inserted in inversion.
