山梨酸钾在红豆杉开放式组织培养中的应用

目的]确定山梨酸钾作为开放式组织培养抑菌剂的最佳浓度。方法]通过在培养基中添加不同浓度的山梨酸钾代替高压灭菌，研究了山梨酸钾在红豆杉开放式组织培养中的抗菌作用。结果]0．02和0．04g/L山梨酸钾的抑菌效果较好，抑菌时间最长，在红豆杉的开放式组织培养中未出现青霉和黑曲霉，且菌块生长较慢。在含0．02g／L山梨酸钾的培养基上培养的红豆杉外植体在第16天出现愈伤组织。在含0．02和0．04g/L山梨酸钾的培养基上外植体的存活率分别为13％和3％，其余培养基上的外植体全部死亡。在含0．02和0．04g／L山梨酸钾的培养基上外植体的污染率分别为87％和97％。0．02g／L山梨酸钾的抑菌效果明显好于其他浓度。结论]山梨酸钾可以作为红豆杉开放式组织培养的前期抑菌剂。
