摘    要:[目的]寻找一种有效的泥火山土壤宏基因组DNA的提取方法。[方法]分别采用CTAB法、SDS加酶法、实验室改进法、试剂盒法提取泥火山土壤宏基因组,比较4种方法的提取效果。[结果]试剂盒法的提取效率最高、纯度最好;实验室改进法次之;SDS加酶法提取量最多。实验室改进法获得的DNA稀释10倍可直接用于PCR扩增。[结论]从经济和DNA质量的角度考虑,实验室改进法可作为新疆泥火山土壤宏基因组DNA提取最有效的方法。

Study on Extraction Method of Metagenomic DNA from Mud Volcano in Xinjiang

Abstract:[Objective] The aim was to seek an effective extraction method of metagenomic DNA from Mud Volcano. [Method]The metagenomic DNA from Mud Volcano was extracted by CTAB extraction method,SDS-enzyme method,improved method,reagent sets method,respectively. The extraction of four kinds of methods were compared. [Result] The extracted rate and purity in reagent sets method were the highest,next was improved method,the extracted quantity in SDS-enzyme method was maximum. Ten times diluted DNA which extracted by the improved method could be used for PCR. [Conclusion]Considering economy and DNA quality,the improved method could be used as an effective extraction method of metagenomic DNA from Mud Volcano.
