摘    要:应用基因枪法与农杆菌介导法将来源于山菠菜的BADH(betaine aldehyde dehydrogenase)基因导入水稻。从获得转基因植株数量、转化率、转化植株PCR阳性率、外源基因拷贝数及其表达活性、T0代植株的结实率对两种转化方法进行了比较。结果表明,用农杆菌介导法转化BADH基因的转化效率和PCR阳性率比基因枪法更高,且用农杆菌介导法得到的转基因植株T0的结实率等农艺性状也优于基因枪法。

Abstract:The transformation effects of BADH gene (encod ing betaine aldehyde dehydrogenase) into rice by particle bombarment and Agr obacterium tumefaciens mediated were compared in transgenic plant number, t ransformation efficiency, rate of PCR positive plantlets, foreign gene copies and their expression activity, and seed setting rate of T 0 generation. Resu lts showed that transformation efficiency and rate of PCR positive plantlets by Agrobacterium tumefaciens mediated were higher than those by particle bombardment method. Moreover, the performance in seed setting rate of T 0 ge neration and some other agronomic traits of transgenic rice obtained from A grobacterium tumefaciens mediated gene transfer system were better compared w ith particle bombarment methodology.
