珊瑚状猴头菌多糖对大鼠肝抗氧化及代谢调节

摘    要:探讨珊瑚状猴头菌多糖的抗氧化作用和对大鼠肝脏代谢的相关基因表达的影响。将大鼠随机分为正常对照组、多糖干预组和模型组。对照组喂饲基础饲料,多糖干预组(剂量1组、剂量2组)和模型组喂饲高胆固醇饲料,同时多糖干预组(剂量1组、剂量2组)给予多糖灌胃5周,5周后处死大鼠取肝脏。分别测定大鼠肝脏的丙二醛(MDA)含量,总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)的活力。使用实时荧光定量PCR分别检测大鼠肝脏相关基因表达量:酰辅酶A:胆固醇酰基转移酶2(ACAT2)、卵磷脂胆固醇酰基转移酶(LCAT)。与模型组比较:多糖剂量1组、剂量2组大鼠肝脏的MDA含量显著下降(P〈0.01),T-SOD活力显著升高(P〈0.01),GSH-Px活力显著升高(P〈0.01),CAT活力显著提高(P〈0.01);多糖剂量1组、多糖剂量2组大鼠肝脏的基因表达量ACAT2mRNA显著下降(P〈0.01);LCATmRNA显著升高(P〈0.01)。珊瑚状猴头菌多糖具有显著的抗氧化作用,对大鼠肝脏的代谢具有调节作用。

Abstract:The effect of antioxidant and the expression of related genes in rat liver metabolism of Hericium coralloides polysaccharide were explored. The rats were randomly divided into control group, intervention group and control group of polysaccharides. The control group was fed with basic feed, polysaccharide intervention group (dose group 1, dose group 2) and model group fed with high choles terol diet, and polysaceharide intervention group (dose group 1, dose group 2) gave polysaceharide orally for 5 weeks, the rats were killed after 5 weeks of livers. Rat liver MDA content, superoxide dismutase (T - SOD), glutathione peroxidase ( GSH - Px), catalase (CAT) activity were measured. Real -time fluorescence quantitative PCR were used to detect the gene expression related to rat liver index: A acetyl CoA: cholesterol acyltransferase2 (ACAT2), lecithin cholesterol acyhransferase (LCAT). The resuhs showed that compared with the model group, polysaceharide dose group 1 and group 2 doses of MDA content significantly decreased in rat liver(p 〈 0. 01 ), T- SOD activity increased significantly (p 〈0. 01 ), GSH- Px activity increased significantly (p 〈0.01 ), and CAT activity was significantly improved (p 〈0. 01 ). Liver gene expression index of polysaccharide dose group 1 and group 2, ACAT2 mRNA de- creased significantly(p 〈0. 01 ), and LCAT rnRNA increased significantly (p 〈0. 01 ). The Hericium coralloides polysaccharide had sig- nificant antioxidant effect, which had regulatory effect on metabolism in rat liver.
